Will any quests fail if I go to Skellige?

No. Once you’ve been there, you can return to Velen from Skellige via fast travel. The only thing that auto-fails quests is progressing through certain main story quests.

You can complete the Skellige questline, and most of the Kaer Morhen questline before Now or Never will become unavailable. The game will prompt you to do Now or Never when you get to a quest called Brothers In Arms, and if you don’t do it then, it will fail.
